Sequence of amino acids in polypeptide chain that is what we call primary structure. And this sequence is governed by the nature of side chains(R) of each amino acid.

Explanation:

Enzymes(except ribozyme) are actually protein molecules. And the sequence of amino acid in each enzyme is distinct and it is something that determines the specific three-dimensional shape of an enzyme.
Enzymes being protein molecules are folded in very precise shapes. Changing the order of amino acid would cause it to fold into a different shape. Consequently, would change or distort the shape of the enzyme.
And hence, the specific catalytic property of enzyme would also be diminished.
