The slope is (change in $y$) / (change in $x$).
The change in $y$ is the difference of the $y$ coordinates and the change in $x$ is the difference of the $x$ coordinates:
slope $= \frac{\Delta y}{\Delta x} = \frac{- 3 - \left(- 2\right)}{\frac{1}{4} - \frac{3}{4}}$
$= \frac{- 3 + 2}{\frac{1 - 3}{4}} = \frac{- 1}{- \frac{2}{4}} = \frac{- 1}{- \frac{1}{2}} = 2$
