When the replication of chromosomes is not followed by karyokinesis and cytokinesis, it leads to endomitosis and the nucleus contains multiple sets of chromosomes.

Explanation:

Endomitosis is a type of mitosis in which mitosis is initiated but does not get completed.
For example, the alkaloid Colchicine inhibits the formation of mitotic spindle and hence the cells get stuck at metaphase.
In this case, the chromosomes have already replicated ( in S phase) but the cell is not able to complete it's cycle. Hence, multiple sets of chromosomes are found in the cell which is why it is called a polyploid cell.
