About 167 million years ago.

Explanation:

The oldest fossil of a reptile with anatomical similarities with those of a today's snake date back to about 167 million years ago and it was found in a quarry near Oxford, UK and it was about 25cm long.
The second oldest snake fossil is dated 102 million years ago.
Both of the above specimens had limbs (however very small) but scientists believe that they served only for a better grasp when crawling rather than a tool for walking.
