DNA is a double helical molecule. During both replication and transcription, the helix opens up.

During replication , two exact copies are made from original DNA. Both strands of DNA act as template during replication, which is a semi-conservative process, i.e. in the daughter DNA, one parental strand is retained and a new complementary strand is synthesised.

During transcription of RNA, only one DNA strand will act as template; the other strand is called coding strand. Thus the coding strand actually does not participate in transcription process.
