How do we know our galaxy is rotating?

We don't quite know yet. There are only mere speculations.

Explanation:

Scientists and Astronomists have not yet found an absolute way of identifying whether or not our Galaxy is at all, spinning on an axis or on different axies.

Theorists believe that there is another type of energy causing our Galaxy to move in a fashion, fooling us humans into believing that we are indeed revolving around an axis.
Others believe our Galaxy isn't moving at all due to the lack of Gravitational Energy and Conservation of Angular Momentum in our Solar System (due to stability?).
