You dilute the stuff $\text{1000-fold}$.
And thus take $1 \cdot m L$ of the original sample, place it a $1 \cdot L$ volumetric flask, and dilute to volume.....i.e.
(1*mLxx4.3xx10^4*"cells"*mL^-1)/(1000*mL)=43*"cells"*mL^-1.
